Senior Research Associate - Muscle Program Team. The Gene Therapy Group at Dyno applies technologies created by the Dyno platform to discover novel AAV capsids with the potential to improve gene therapies. The group is organized into Program Teams, each with a focus on improving delivery to a target tissue type. The Muscle Program is applying the Dyno platform toward improving AAV for delivery of gene therapies to skeletal muscle. We are building a team of researchers motivated by an interest in muscle biology and a drive to improve treatment options for severe muscle diseases. This role requires a broad molecular biology skillset and the ability to perform a variety of assays using standardized protocols. You will learn and apply diverse methods for building libraries of AAVs and characterizing their properties in muscle. You will also work to optimize AAV engineering protocols for application to muscle. This is a highly collaborative position in which you will integrate across the many disciplines represented within Dyno, including cloning, tissue processing, vector production, and in vitro assays. While your primary focus will be on muscle, you will have opportunities to learn from and contribute to a diverse set of scientific and cultural projects.
Build AAV vector constructs and produce AAV vectors
Conduct assays in cultured muscle-derived mammalian cell lines
Extract and quantify nucleic acid and protein from mammalian cells and tissues
Implement next-gen sequencing to measure AAV properties
Follow established lab protocols and contribute to new experimental method development
Collect, analyze, and present assay results
Successfully collaborate with colleagues across teams
Assist in lab organization, including ordering supplies, restocking, and sample tracking

BS/MS or MS in biology, biochemistry, biomedical engineering, or related fields
3+ years hands-on wet lab experience, preferably within the biotech industry
Experience in muscle tissue handling, culturing muscle-derived mammalian cells, and aseptic technique
Experience in DNA/RNA purification and analysis protocols (qPCR, RT-qPCR, ddPCR, etc.)
Hands on molecular cloning experience
Ability to follow advanced protocols and procedures
Ability to document and maintain an organized account of lab work and results
Skilled in Excel/Prism, Word, PowerPoint, and other relevant software
Experience working with high-throughput assays, e.g., automated sample processing, multi-channel pipetting, molecular barcoding
Experience in primary culture of mammalian cardiac and/or skeletal muscle cells or organoids
Experience in recombinant AAV production and purification
Experience in sequence design and analysis software, e.g., BLAST, Benchling, etc.
NGS library preparation and sequencing on Illumina platforms
